SRA Antibody (D-8) is a mouse monoclonal IgM antibody that detects steroid receptor RNA activator (SRA) in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). SRA plays a crucial role in mediating the transactivation of steroid hormone receptors, functioning as both an RNA transcript and a stably expressed protein that forms complexes with steroid receptor coactivator-1. SRA contains six essential RNA motifs that are vital for coactivation capabilities, underscoring its importance in regulating gene expression in response to steroid hormones. SRA is widely expressed across various normal tissues, with particularly high levels found in the liver and skeletal muscle, while exhibiting lower expression in the brain. SRA levels are elevated in breast tumors compared to normal tissue, suggesting a potential role in cancer biology. Overexpression of SRA enhances estrogen receptor alpha transcriptional activity, and when cells are transfected with antisense oligodeoxynucleotides targeting SRA, a dose-dependent reduction in estrogen receptor alpha expression occurs. SMRT/HDAC1 associated repressor protein (SHARP) interacts with SRA, inhibiting SRA′s ability to potentiate steroid receptor transcription, further highlighting the complex regulatory mechanisms involved.
