SPAG5 Antibody (LX-8) is a mouse monoclonal IgG3 kappa light chain antibody that detects the SPAG5 protein of human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Anti-SPAG5 antibody (LX-8) is available as the non-conjugated format. SPAG5, also known as sperm associated antigen 5, MAP126, hMAP126, DEEPEST, or Astrin, is a 1,193 amino acid protein predominantly expressed in the testis, with lower expression levels found in the liver, placenta, pancreas, colon, and thymus. SPAG5 protein is critically localized to spindle poles during prophase and throughout the spindle during metaphase and anaphase, making SPAG5 essential for proper spindle formation. SPAG5 plays a crucial role in the dynamic regulation of mitotic spindles, which is essential for accurate chromosome segregation during cell division. Additionally, SPAG5 is involved in sister chromatid cohesion and the subsequent separation of chromatids, thereby serving as a vital regulator of chromosome integrity. SPAG5 features a long stalk and a globular head domain that allows SPAG5 to form a homodimer characterized by aster-like structures, which are essential for its function. Notably, upon DNA damage, SPAG5 may undergo phosphorylation by ataxia-telangiectasia mutated (ATM) or ataxia telangiectasia and Rad3-related (ATR) kinases, highlighting SPAG5′s involvement in cellular responses to genomic stress.
