SP-100 Antibody (1G6) is a mouse monoclonal IgG2a kappa light chain antibody that detects SP-100 of human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). SP-100 (1G6) antibody is available as non-conjugated anti-SP-100 antibody. The human SP100 gene encodes an autoantigen that co-localizes with PML and NDP52 in distinct nuclear domains known as nuclear dots (NDs) or ND10 nuclear bodies, which are crucial for various cellular processes. These nuclear domains serve as sites where papova-, adeno-, and herpesviruses initiate transcription and DNA replication, highlighting their significance in autoimmunity, viral infections, and the development of acute promyelocytic leukemia. SP-100 is an interferon-inducible protein that exists in two splice variants, one containing a highly conserved sequence from the DNA-binding high mobility group 1 protein, representing a novel HMG-box protein. This alternatively spliced variant exhibits a unique expression and localization pattern differing from full-length SP-100. SP-100 undergoes post-translational modification by small ubiquitin-related modifier SUMO-1, essential for functional activity. SP-100 features a functional nuclear localization signal and an ND-targeting region overlapping with a homodimerization domain, referred to as HSR domain, considered a novel protein motif. SP-100 interacts with Bright, a B cell regulator of immunoglobulin heavy chain transcription, and associates with LYSP100/SP140, the lymphoid-restricted homolog of SP100, underscoring a role in immune regulation.
