SOX4 encodes the transcription factor Sox-4, a high-mobility group (HMG) DNA-binding protein that regulates lineage specification, differentiation, and cell fate decisions during development and tissue homeostasis. Sox-4 participates in transcriptional programs linked to Wnt/β-catenin and TGF-β/SMAD signaling, and it can influence epithelial–mesenchymal transition, cell-cycle control, and apoptosis depending on cellular context. Altered SOX4 expression has been associated with dysregulated progenitor maintenance and aberrant transcriptional networks observed across multiple cancer and immunologic research settings. As a nodal regulator of gene expression, Sox-4 is frequently studied for its effects on chromatin-associated transcriptional circuitry and pathway cross-talk.
Sox-4 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the SOX4 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the SOX4 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the SOX4 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Sox-4 protein expression.
This CRISPR knockout system enables efficient generation of SOX4-deficient cell models for investigation of Sox-4 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
