SEE ALSO...

Ski Antibody (G8) is a mouse monoclonal IgG1 kappa light chain antibody that detects the Ski protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). Anti-Ski antibody (G8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The Ski protein plays a crucial role in cellular processes as a transcriptional regulator, particularly in the context of TGFβ signaling pathways. Ski protein is primarily located in the nucleus, where Ski protein interacts with Smad proteins to modulate gene expression. This nuclear localization is vital because Ski protein effectively represses Smad-mediated transcriptional activation, thereby inhibiting TGFβ-induced cell growth and extracellular matrix production. Ski protein contains five tandem repeats in its C-terminal domain and two leucine zipper motifs, which are essential for DNA binding capabilities and the formation of homodimers and heterodimers. By regulating the activity of Smad proteins, Ski monoclonal antibody (G8) provides valuable insights into the mechanisms of oncogenesis and cellular transformation, making anti-Ski antibody (G8) an important tool for researchers studying cancer biology and related fields.

When should the TransCruz reagent sc-33693 X be used instead of the standard form sc-33693?

Asked by: Randy McDonald
Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.

Rated 2 out of 5 by from Western blottingA strong single band was detected in 293T cells overexpressed human Ski protein. We couldn't detect endogenous human Ski protein using 1:1000.

Rated 4 out of 5 by from Works wellWe just purchased SKi for the lab. Using 20 ug protein, ski at 1:500 dilution, and supersignal ECL, we see defined bands around 100 kDa. There is some nonspecific binding so I rated 4 out of 5 stars
