How do I modify this kit/protocol to use for electroporation of sirna into oragnoids using a amaxa nucleofector 2B device with cuvettes of 1mm gap?

Thank you for your question. Please refer to the specifications of your electroporation kit and machine. Optimum parameters for highest electroporation rate also depend on the particular samples you are working with. We do not have any data on parameters for transfection organoids.

Thank you for your question. In our siRNA protocol, titled "siRNA mediated inhibition of gene expression", for the siRNA duplex, we are referring specifically to the two strands, sense strand and anti-sense strand, of the siRNA reagent.

Works well!I bought siRNA Reagent System: sc-45064 a months ago. I used the siRNA reagent system to knock down expression of the PGC-1α protein in rat aortic endothelial cells. Transfection of the rat PGC-1α siRNA (sc-72151) and negative control siRNA was carried out according to kit instructions. It was very easy to follow instructions and worked very well.

Worked on the first try!I used the siRNA reagent system to knock down expression of the RWDD4 protein in A172 glioblastoma cells. Transfection of the human RWDD4 siRNA (sc-88908) and negative control siRNA was carried out according to kit instructions, but scaled down for a 12 well plate. After 48 hours, cell lysates were prepared and blotted via standard western blotting techniques. As can be seen in the Figure, expression of the ~25 kDa band for RWDD4 was greatly reduced, which GAPDH (37 kDa band) was unaffected.
