SGI-1027 is a potent inhibitor of DNA methyltransferases (Dnmt1, Dnmt3a, and Dnmt3b) that play roles in DNA methylation. By blocking the activity of DNA methyltransferase 1 (DNMT1) and inducing its degradation, SGI-1027 exhibits the ability to reactivate silenced tumor suppressor genes by reducing CpG island hypermethylation. The selective degradation of DNMT1 was observed in various cancer cell lines upon treatment with SGI-1027, while DNMT3A and DNMT3B remained minimally affected. Within 24 hours, at concentrations comparable to decitabine, complete degradation of DNMT1 protein was achieved without significantly impacting its mRNA level. These findings highlight the potential of SGI-1027 in epigenetic cancer therapy as it holds promise for selectively reactivating tumor suppressor genes through its potent inhibition of DNMTs.
