Separase Antibody (B-4) is a mouse monoclonal IgG2a kappa light chain antibody that detects Separase protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-Separase antibody (B-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Separase is a crucial cysteine protease that plays a vital role in regulating cell cycle, specifically in triggering anaphase by facilitating sister chromatid separation during mitosis. Separase maintains genomic stability by ensuring each daughter cell receives an accurate complement of chromosomes. Separase achieves this by hydrolyzing the SCC1 subunit of cohesin, a protein complex that holds sister chromatids together. During non-division phases, Separase remains inhibited through association with securin. Upon anaphase onset, securin undergoes degradation by the anaphase-promoting complex/cyclosome, releasing active Separase. Separase also activates transiently during meiosis and may contribute to homologous chromosome separation, highlighting a significant role in both mitotic and meiotic cell divisions.

What are the optimal western conditions for separase? overnight transfer at low voltage? PVDF membrane? low % gel? Thanks!

Thank you for your question. We recommend using an 8% gel, and a wet transfer to PVDF membrane 100 mAmps (0.1 Amp) overnight at 4C.

What is the recommended fixation method for immunofluorescence staining with Separase (B-4): sc-390314 monoclonal antibody?
