Salinomycin is a polyether antibiotic that functions as an ionophore, facilitating the transport of monovalent cations across cell membranes. It acts as a potassium ionophore, disrupting the ion balance within cells. This disruption leads to the depolarization of mitochondrial membranes, ultimately inhibiting oxidative phosphorylation and ATP production. Salinomycin may induce apoptosis in cancer cells by interfering with various cellular processes, including the Wnt/β-catenin signaling pathway. This compound may inhibit the activity of ATP-binding cassette (ABC) transporters, which play a role in multidrug resistance in cancer cells. Salinomycin′s mode of action involves disrupting ion balance and mitochondrial function, as well as interfering with cellular signaling pathways and chemical resistance mechanisms.
