SAG is a potent agonist of Hedgehog (Hh) signalling. It operates by activating the Smoothened (Smo) protein function. SAG directly binds to Smo and can block Smo inhibition by cyclopamine. This binding of SAG alters the Smo conformation, causing an accumulation in cilia and activation of gene transcription. The conversion of human induced pluripotent stem fcells to the neural lineage is improved by higher concentrations of SAG (0.4 mM).
