S-antigen Antibody (PFS-3) is a mouse monoclonal IgG1 antibody that detects S-antigen of Plasmodium falciparum by western blotting (WB). S-antigen is a crucial protein in the life cycle of the malaria-causing protozoan parasite, Plasmodium falciparum, and plays a significant role in the parasite′s ability to evade the host′s immune response. Located in the parasitophorous vacuole of the mature asexual intraerythrocytic parasite, S-antigen exhibits considerable antigenic diversity, which poses challenges for vaccine development. The S-antigen gene comprises multiple alleles originating from the same chromosomal site, with each allele featuring a central region of tandemly arranged, nearly identical peptides that are specific to that allele. This structural characteristic allows for the identification of distinct serotypes based on the repeat region, while the flanking non-repetitive sequences define four different S-antigen families, each exhibiting significant variation, particularly in the C-terminal-flanking region. The ability to differentiate between these families is essential for understanding the immune response to malaria and developing effective therapeutic strategies.
