Ru360, an oxygen-bridged dinuclear ruthenium amine complex, has been noted to selectively and potently inhibit mitochondrial calcium uptake. It is cell-permeable. Exposure of mitochondria to Ru360 in vitro produced no effect on actomyosin ATPase activity, sarcolemmal Na+/Ca2+ exchange, sarcoplasmic reticulum Ca2+ uptake or release, L-type Ca2+ channel current, or cytosolic Ca2+ transients. These results indicate that Ru360 only affects calcium uniporters in the mitochondria. It does not compete with ATP.
