RPMI 1640 Medium was first developed in 1966 at Roswell Park Memorial Institute by Moore and his colleagues. It was originally designed as a variation of McCoy′s 5A Medium to support lymphoblastoid cells in suspension culture. However, it was later found to be effective for a broad range of anchorage-dependent cell types. Initially formulated for use with a serum supplement, RPMI 1640 has also demonstrated the ability to sustain certain cell lines without serum. Additionally, it has been extensively utilized in hybrid cell growth and fusion protocols. This medium is well-suited for culturing both normal and neoplastic human leukocytes.
