Rin3 encodes RIN3, a Ras and Rab interactor that functions as a guanine nucleotide exchange factor for Rab5 family GTPases, linking receptor tyrosine kinase signaling to endocytic trafficking. In mouse cells, RIN3 participates in clathrin-mediated endocytosis, early endosome maturation, and regulation of receptor internalization and recycling, processes that shape downstream signaling amplitude and duration. Through these roles, Rin3 influences vesicle dynamics, membrane turnover, and cytoskeletal coordination during cell activation states. Altered RIN3 activity has been connected to disease-relevant mechanisms involving dysregulated endosomal trafficking and receptor signaling, supporting its study in pathways that contribute to inflammatory and neurodegenerative phenotypes.
RIN3 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Rin3 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Rin3 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Rin3 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish RIN3 protein expression.
This CRISPR knockout system enables efficient generation of Rin3-deficient cell models for investigation of RIN3 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
