Rho Kinase Inhibitor III, Rockout is a cell permeable ATP-competitive Rock (Rho Kinase) inhibitor discovered by an image based, high throughput screen of a wound healing cell monolayer. When discovered Rock (Rho Kinase) Inhibitor III, Rockout was observed to not be toxic while inhibiting blebbing and promoting cell spreading. The compound was observed to inhibit the blebbing of recently trypsinized Swiss3T3 cells and M2 cells. The Inhibitor was documented to have an IC50 =12 μM and at 50 μM inhibit blebbing in most cells. The concentration of 50μM was observed to lead to the dissolution of cellular stress fibers. Rho Kinase Inhibitor III, Rockout has been observed to be five times less potent than Y-27632.
