RCAN2/3 Antibody (B-9) is a mouse monoclonal IgG2a kappa light chain antibody that detects RCAN2 and RCAN3 proteins of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). RCAN2/3 (B-9) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. RCAN2, also known as Calcipressin-2 or thyroid hormone-responsive protein ZAKI-4, is a 197 amino acid protein that plays a crucial role in regulating calcineurin-dependent transcriptional responses by binding to calcineurin A′s catalytic domain. This interaction helps modulate calcium signaling pathways, vital for various cellular processes, including immune response and neuronal function. RCAN2 is constitutively expressed in endothelial cells and inhibits calcineurin activity, thereby restraining vascular endothelial growth factor-mediated angiogenesis, essential for proper blood vessel formation and repair. RCAN2 expression occurs in fibroblasts, heart, brain, liver, and skeletal muscle, but not in placenta, lung, kidney, and pancreas, with expression being upregulated by physiological concentrations of triiodothyronine. RCAN3, or Calcipressin-3, is a 241 amino acid protein that also interacts with calcineurin and is implicated in central nervous system development. RCAN3 overexpression inhibits calcineurin activity towards nuclear factor of activated T-cells (NFAT) transcription factors, thereby downregulating NFAT-dependent cytokine gene expression in activated Jurkat T-cells. RCAN3 shows low expression in various tissues but high expression in kidney, heart, liver, peripheral blood lymphocytes, and skeletal muscle, and interacts with cardiac troponin I, suggesting a potential role in cardiac contraction events.
