rac-Etomoxir is a potent inhibitor of carnitine palmitoyltransferase A (CPT1), an enzyme for the oxidation of long-chain acyl CoA esters. Specifically, it acts as a strong inhibitor of mitochondrial CPT1. Etomoxir′s main mode of action involves blocking CPT1, thereby impeding the transport of long-chain fatty acids into the mitochondria for oxidation. Extensive obesity and diabetes studies have been conducted with etomoxir. These studies have also explored the impact of etomoxir inhibition on cardiac function, energy metabolism, and muscle metabolism in vitro. Additionally, etomoxir has been utilized to examine its role in regulating hepatic fatty acid oxidation and glucose metabolism. By binding to CPT1 and inhibiting its activity, etomoxir effectively disrupts the transport of long-chain fatty acids into the mitochondria for oxidation. This inhibition leads to an elevation in glucose utilization, resulting in reduced production of reactive oxygen species and improved insulin sensitivity.
