rabphilin-3A Antibody (D-6) is a mouse monoclonal IgG2a kappa light chain antibody that detects rabphilin-3A of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-rabphilin-3A antibody (D-6) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. rabphilin-3A plays a crucial role in calcium-dependent exocytosis, acting as a positive regulator in this process. rabphilin-3A is primarily located in neuroendocrine cells, where rabphilin-3A co-localizes with Rab3A on synaptic vesicles and chromaffin granules, highlighting rabphilin-3A′s importance in neurotransmitter release and hormone secretion. rabphilin-3A′s structure includes two C2 domains at the carboxy-terminal and a cysteine-rich region with two zinc finger motifs at the amino-terminal, which are essential for interaction with calcium ions and phosphatidylinositol 4,5-bisphosphate-containing lipid vesicles. These structural features enable rabphilin-3A to bind to Rab3A in a GTP/Mg²⁺-dependent manner, facilitating the regulation of exocytosis. While rabphilin-3A and Rab3A interact, both appear to influence exocytosis independently, suggesting that rabphilin-3A may mediate effects through interactions with an unidentified factor that recognizes the Rab3 binding domain. rabphilin-3A monoclonal antibody (D-6) serves as an invaluable tool for researchers studying the mechanisms of exocytosis and cellular signaling pathways.
