(R)-Hydroxy Des(boric acid) Bortezomib is an impurity in the synthesis of Bortezomib. It lacks the boric acid moiety that is present in the parent compound. A reversible inhibitor of the 26S proteasome-a barrel-shaped multiprotein particle found in the nucleus and cytosol of all eukaryotic cells. Targets the ubiquitin-proteasome pathway. The inhibition of this proteasomal activity leads to the accumulation of defective proteins in cells, causing cell cycle arrest and apoptosis. By comparing the activity of bortezomib with its des(boric acid) counterpart, the role of the boric acid group in the mechanism of action and its interactions with the proteasome can be better understood.
