PYGO1 encodes Pygopus 1, a nuclear co-activator that couples β-catenin/TCF transcriptional complexes to chromatin in canonical Wnt signaling. Through interactions with BCL9/9L and histone-modifying machinery, Pygopus 1 supports transcriptional programs controlling cell fate specification, proliferation, and epithelial homeostasis. PYGO1-dependent Wnt outputs intersect with processes such as stem-like maintenance and differentiation, and altered regulation of this pathway is frequently implicated in oncogenic transcriptional states and developmental dysregulation. These features make PYGO1 a useful target for dissecting Wnt-driven gene expression and chromatin engagement in human cellular models.
Pygopus 1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the PYGO1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the PYGO1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the PYGO1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Pygopus 1 protein expression.
This CRISPR knockout system enables efficient generation of PYGO1-deficient cell models for investigation of Pygopus 1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
