

PSGL-1 Antibody (215) is a mouse monoclonal IgG2a antibody that detects PSGL-1 in human samples through flow cytometry (FCM). PSGL-1, also known as P-Selectin glycoprotein ligand or CD162, plays a crucial role in the immune response by mediating leukocyte adhesion to the endothelium during inflammation. PSGL-1 is primarily located on leukocyte microvilli tips, where a unique structure, characterized by a disulfide-linked homodimer and a rich extracellular domain composed of serines, threonines, and prolines, facilitates selectin interactions. The presence of 15 and 16 decameric repeats in HL-60 and U-937 cells, as well as in human leukocytes, enhances binding affinity to P-Selectin and E-Selectin, which is essential for leukocyte trafficking and recruitment to inflammation sites. For optimal selectin binding, PSGL-1 must undergo specific post-translational modifications, including sialylation and fucosylation, which are critical for function as a cell adhesion receptor. PSGL-1 belongs to a broader family of sialomucins, which transduce negative signals in hematopoietic cells, thereby influencing immune responses.
