POH1 Antibody (SQ-17) is a mouse monoclonal IgG1 kappa light chain antibody that detects POH1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). POH1 (SQ-17) antibody is available as the non-conjugated form. POH1, a human homolog of the yeast protein Pad1, plays a crucial role in the 26S proteasome, which is essential for the degradation of proteins tagged for destruction via the ubiquitin pathway. Specifically, POH1 is a component of the 19S regulatory cap of the 26S proteasome, where POH1 functions as a deubiquitinase, removing ubiquitin chains from substrates and facilitating their entry into the proteasome′s catalytic core. This process is vital for maintaining cellular protein homeostasis and regulating various cellular functions, including cell cycle progression and stress responses. POH1 is predominantly expressed in heart and skeletal muscle tissues, highlighting POH1′s importance in muscle function and health. Notably, transient overexpression of POH1 in mammalian cells has been shown to confer resistance to chemotherapeutic agents such as taxol and doxorubicin, indicating POH1′s potential role in cancer biology and therapeutic resistance. The gene encoding human POH1 is located on chromosome 2q24.2, and interestingly, the Pad1 homolog found in marine sponges (POHL) is upregulated in response to toxic compounds like staurosporine and taxol. In the parasitic organism Schistosoma mansoni, the Pad1 homolog SmPOH appears to stabilize the transcription factor c-Jun, enhancing c-Jun levels for the transactivation of AP-1-responsive genes, further underscoring the diverse biological significance of POH1 across different species.
