PKR Antibody (FJ-6) is a mouse monoclonal IgG2a antibody that detects PKR in human samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). PKR, an interferon-inducible, RNA-dependent protein serine/threonine kinase, plays a crucial role in cellular response to viral infections by phosphorylating the α subunit of protein synthesis initiation factor eIF-2. This phosphorylation event on serine-51 leads to translation inhibition, effectively halting viral replication and allowing host cells to mount an immune response. PKR′s structure is characterized by distinct functional domains, with serine/threonine kinase catalytic domains located in the carboxy terminal half and RNA binding domains situated in the amino terminal region. This structural organization enables PKR to respond to various stimuli, including double-stranded RNA, a hallmark of viral infection. PKR′s enzymatic activity is finely tuned through multiple regulatory mechanisms, including transcriptional regulation in response to interferon, autoregulatory mechanism at the translational level, and post-translational modifications such as RNA-mediated autophosphorylation. Anti-PKR antibody (FJ-6) serves as an essential tool for studying immune response and viral pathogenesis pathways.
