Pit-1 Antibody (G-2) is a mouse monoclonal IgG2a kappa light chain antibody that detects Pit-1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Pit-1 (G-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Pit-1, also known as growth hormone factor-1 (GHF-1), is a crucial transcriptional regulator that plays a significant role in anterior pituitary gland development, where GHF-1 is essential for somatotrope, lactotrope, and thyrotrope differentiation and function. Pit-1 is characterized by a POU domain, containing a bipartite DNA-binding domain that interacts with specific gene promoters, activating growth hormone and prolactin transcription. Pit-1′s dual role as an activator and repressor of gene expression maintains hormone production balance and ensures proper pituitary function. Mutations in Pit-1 can lead to combined pituitary hormone deficiency (CPHD), affecting growth hormone, prolactin, and thyroid-stimulating hormone secretion, underscoring Pit-1′s importance in endocrine health. Pit-1′s gene location on human chromosome 3p11 highlights evolutionary conservation and significance across species.

Can Pit-1 Antibody (G-2): sc-25258 X detect chicken in ChIP?

Asked by: Ellyse
Thank you for your question. Unfortunately this antibody has not been epitope mapped, so I am not sure if it can or can not detect chicken Pit-1. However, we would be happy to send you a sample of this antibody, sc-25258 if you would like to try it on your chicken samples! Please contact your local distributor, they will be happy to help get you the sample. You can find our distributor contact information here: https://www.scbt.com/scbt/resources/distributors

When should the TransCruz reagent sc-25258 X be used instead of the standard form sc-25258?

Asked by: Dr Ninau Qelp
Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.
