PARP-10 Antibody (5H11) is a rat monoclonal IgG1 antibody raised against human PARP-10, with reactivity in mouse, rat, and human samples, making anti-PARP-10 antibody (5H11) suitable for western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF) applications. PARP-10, also called ARTD10 and PARP10, is a member of the poly(ADP-ribose) polymerase family, involved in regulating cell proliferation and cellular transformation. Unlike nuclear counterparts, PARP-10 exists in both nucleus and cytoplasm, playing a distinct role in controlling oncogenic signaling pathways. One key interaction of PARP-10 is with oncogene c-Myc, a transcription factor regulating cell growth and division. PARP-10 inhibits c-Myc-mediated transformation by adding ADP-ribose units to target proteins, modulating their activity. Additionally, PARP-10 interacts with E1A, another protein involved in cell cycle control, further contributing to a role in suppressing oncogenic transformation. These interactions highlight PARP-10′s critical function in maintaining cellular homeostasis and preventing unchecked cell proliferation. PARP-10 (5H11) antibody′s ability to detect PARP-10 in various species and experimental setups makes anti-PARP-10 antibody (5H11) a valuable tool for studying regulatory pathways and mechanisms of cell growth inhibition associated with PARP-10.

I am using PARP-10 (5H11): sc-53858 for immunofluorescence, what dilution of this antibody should I use?

Asked by: Professor Griffin
Thank you for your question. We recommend a starting dilution of 1:50. However, optimal antibody concentration should be determined by titration. Please find our full immunofluorescence protocol here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
