P5CRL Antibody (E-63) is a mouse monoclonal IgG2a kappa light chain antibody that detects P5CRL protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-P5CRL antibody (E-63) is available as a non-conjugated format. P5CRL protein, also known as pyrroline-5-carboxylate reductase-like, is crucial in proline biosynthesis, a process vital for maintaining cellular homeostasis and responding to stress. Proline plays a significant role in protein folding, stability, and structure, and its presence can act as a sequence-recognition motif in various proteins. Additionally, proline functions as a non-enzymatic antioxidant, helping to mitigate damage from reactive oxygen species (ROS) across different organisms, including microorganisms, animals, and plants. P5CRL protein consists of 274 amino acids and belongs to the pyrroline-5-carboxylate reductase family, functioning as a homodecamer formed by five homodimers. This structural arrangement is essential for enzymatic activity, as P5CRL allows for efficient catalysis in the final step of proline biosynthesis, where pyrroline-5-carboxylate reductase (P5CR) reduces aldehyde dehydrogenase 4A1 (ALDH4A1) to proline using NAD(P)H as a cofactor. P5CRL′s participation in these critical biochemical pathways underscores its importance in cellular function and stress response, making P5CRL monoclonal antibody (E-63) a valuable tool for researchers studying proline metabolism and its implications in health and disease.

I am using P5CRL (E-63): sc-100487 for immunofluorescence, what dilution of this antibody should I use?

    Asked by: jenniferc
Thank you for your question. We recommend a starting dilution of 1:50. However, optimal antibody concentration should be determined by titration. Please find our full immunofluorescence protocol here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
