p38 MAP Kinase Inhibitor VIII is a synthetic 4-aminobenzophenone small molecule identified in an optimization study for inhibitors of p38 MAP kinase. p38 MAP Kinase Inhibitor VIII is described to inhibit the p38alpha and p38beta isoforms of the kinase by occupying the ATP-binding cleft. This inhibition is described to potently block the release of TNF-alpha (IC50 = 5 nM) and the proinflammatory interleukins IL-1beta, IL-6, IL-8, and IL-10 (IC50s = 30, 17, 4, 74 nM). This compound is a highly effective inhibitor that targets the ATP-binding pocket of p38α and p38β2. With just 1 µM of the inhibitor, it achieves substantial inhibition rates of 82% and 93% against p38α and p38β2, respectively. The IC50 value for p38α inhibition is an impressive 40 nM. In addition, this compound shows minimal activity against a range of 57 other kinases, including p38γ, p38δ, Erk1/2, and JNK1.
