Is there any possibility that p-Ser Antibody (16B4): sc-81514 does work for yeast samples as well...?

Thank you for your question. This antibody has only been tested in samples of mouse, rat, human and canine origin. It has not been tested in yeast species.

Hello, Which buffer is better to detect p-Ser antibody, skim-milk or 3-5% BSA? I immunoprecipitated phospho-protein and used 3% BSA in TBS-T buffer for blocking and 1st antibody but I couldn't get specific phosphorylation band.

HI , I recently purchased this pserine antibody and I do see a band at 20kDa. why do i see a band arounf 20kda for pSerine?

As p-Ser Antibody (16B4): sc-81514 detects a broad range of serine-phosphorylated proteins in crude cell extracts, a signal at 20 kDa can respond to a large variety of different proteins of this molecular weight. The identity of this protein could be determined by assays such as MALDI-TOF.

The p-Ser Antibody (16B4) antibody is a monoclonal mouse IgM κ (kappa light chain). Still, you recommend the m-IgGκ BP-HRP: sc-516102 as a secondary antibody. But this a IgG-kappa specific ab. Would it still work?

Just wondering if your products of sc-81516 and sc-81514 recognize any of the phosphorylated S/T/Y, or they require certain residues beside these phosphorylated residues (like a certain motif).

"preferring positively charged amino acids directly neighbored to phosphoserine". Does it mean that phosphoserine without neighboring positively charged amino acids will not be recognized by the 16B4 Ab ?

Thank you for your question. The antibody recognizes a broad range of serine-phosphorylated proteins in crude cell extracts, interacting with pSK and pSP motifs strongly, recognizing substrates of MAP/SAP kinases and CDC kinases. Phosphoserine detection using an antibody generally is affected by the surrounding amino acids sequence, but it is possible that it can detect phosphoserine without neighboring + charged amino acids.
