OTUB1 Antibody (J-61) is a mouse monoclonal IgG2a kappa light chain antibody that detects OTUB1 protein of mouse, rat, and human origin by western blotting (WB) and immunoprecipitation (IP). Anti-OTUB1 antibody (J-61) is available as the non-conjugated form. OTUB1, also known as OTB1, OTU1, HSPC263, or Otubain-1, is a 271 amino acid protein characterized by one OTU (ovarian tumor) domain, classifying OTUB1 within the OTU family of predicted cysteine proteases. OTUB1 functions as a hydrolase that removes ubiquitin residues from target proteins, preventing their degradation and facilitating proper protein turnover. OTUB1 exists in two isoforms, with one isoform being ubiquitous and the other restricted to lymphoid tissues, highlighting specialized roles in different cellular contexts. OTUB1′s regulation of protein degradation is particularly important in immune responses, as OTUB1 interacts with GRAIL, a protein that plays a significant role in T-cell anergy induction. This interaction underscores OTUB1′s importance in modulating immune responses within secondary lymphoid organs, making OTUB1 a vital component in maintaining immune tolerance and preventing autoimmunity.

Thank you for your question. We recommend using 1–10 µl (i.e., 0.2–2 µg) of antibody with 1 ml of cell lysate. However, optimal antibody concentration should be determined by titration. Rated 1 out of 5 by from Band detected isn't OTUB1The band this antibody detects is at the correct weight but it is not OTUB1 since it recognized it in my siRNA knockdown sample. I know the knockdown worked because I was able to confirm it using a different antibody from a different supplier on the same membrane.
