ONX 0914 is an immunoproteasome inhibitor specifically targeting the β5i (LMP7) subunit. Engineered to exhibit potent inhibitory effects on the immunoproteasome while minimizing interactions with the constitutive proteasome, ONX 0914 demonstrates significantly weaker activity towards the latter. By selectively inhibiting the β5i (LMP7) subunit of the immunoproteasome, ONX 0914 effectively disrupts the production of specific MHC class I-restricted T cell epitopes. In addition, it demonstrates weaker activity towards the β5 subunit of the constitutive proteasome. This selective inhibition profile allows ONX 0914 to modulate the immune response without interfering extensively with the proteasomal functions required for normal cellular processes.
