NTH1 Antibody (2660C1a) is a mouse monoclonal IgG1 antibody that detects NTH1 protein of mouse, rat, and human origin by western blotting (WB) and immunoprecipitation (IP). NTH1 (2660C1a) antibody is available as a non-conjugated anti-NTH1 antibody. The human endonuclease III (hNTH1) is a crucial DNA glycosylase that plays a significant role in maintaining genomic integrity by recognizing and excising oxidatively damaged pyrimidines from DNA. hNTH1 is essential for the repair of DNA lesions that can lead to mutations and contribute to various diseases, including cancer. hNTH1 operates through a beta-elimination mechanism, forming a Schiff base with the deoxyribose at the active site, specifically at Lysine-212, after base removal. Notably, full-length human NTH1 is predominantly localized in the nucleus, which is vital for DNA repair, while the mouse variant is primarily found in the mitochondria, with a smaller fraction in the nucleus. This localization difference is attributed to the presence of a nuclear localization sequence in human NTH1 that is absent in the mouse version. Furthermore, NTH1 gene is situated adjacent to the TSC2 gene, which is implicated in tuberous sclerosis, and both genes share a bidirectional promoter essential for transcription. The ability of NTH1 to excise damaged bases underscores its importance in cellular defense against genomic instability, making anti-NTH1 antibody (2660C1a) a valuable tool for researchers studying DNA repair mechanisms and related pathologies.
