NQO1 Antibody (F-8) is a mouse monoclonal IgG2a kappa light chain antibody that detects NQO1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). NQO1 Antibody (F-8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. NQO1, also known as NAD(P)H:quinone oxidoreductase 1, plays a crucial role in cellular defense by catalyzing the reduction of quinones to hydroquinones, thereby detoxifying harmful compounds and protecting cells from oxidative stress, cytotoxicity, and mutagenicity. This enzymatic activity is particularly important in cancer, as many tumors exhibit overexpression of NQO1, which serves as an obligate two-electron reductase that not only deactivates toxins but also activates bioreductive anticancer drugs. NQO1 is a 274 amino acid protein widely expressed across various tissues, although expression levels can vary significantly. NQO1 gene expression regulation is intricately linked to environmental stressors, including xenobiotics, antioxidants, heavy metals, and radiation, with the antioxidant response element (ARE) in the NQO1 promoter being essential for expression. ARE activation by compounds such as tert-butylhydroquinone is mediated through the PI3-kinase pathway, which is upstream of the transcription factor Nrf2. Various transcription factors, including c-Jun, Nrf1, Jun-B, and Jun-D, bind to the ARE to regulate NQO1 expression, while Maf-Maf homodimers and Maf-Nrf2 heterodimers can negatively regulate transcriptional activation.
