Norgestimate is an acetylated progestin that is used together with ethinyl estradiol. It functions by converting into norelgestromin (also known as levonorgestrel 3-oxime), which is the primary active metabolite, and levonorgestrel. The conversion of norgestimate into these compounds takes place in the intestines and the liver through deacetylation. In contrast to levonorgestrel, norgestimate exhibits a strong affinity for the progesterone receptor while displaying minimal androgenic activity. This is achieved through a significant increase in the level of sex hormone-binding globulin (SHBG) induced by the drug. SHBG has a greater binding preference for androgens, thereby inhibiting their effects.
