Nicotinamide 1,N6-ethenoadenine is an analog of beta-NAD which can act as a fluorescent tag in nucleotide binary and ternary complex formation tests. The ADP-ribose moiety from NAD+, Free Acid (sc-208084) and its analogs can be transferred to proteins and protein-bound ADP-ribose by PARP-1 (ADP Poly(ADP-ribosyl) transferase, pADPRT). These studies show that NAD+ is a better substrate in polymer synthesis reactions than Nicotinamide 1,N6-ethenoadenine. In addition, polyacrylamide gels show that proteins bound with ADP-ribose from Nicotinamide 1,N6-ethenoadenine show slower motility through the gel. This study suggests that Nicotinamide 1,N6-ethenoadenine is useful for studying enzymes degrading poly(ADP-ribose) as a result of this compounds fluorescent nature.
