NG2 Antibody (7.1) is a mouse monoclonal IgG1 antibody that detects NG2 in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and flow cytometry (FCM). NG2, also known as melanoma-associated chondroitin sulfate proteoglycan 4, plays a crucial role in stabilizing cell-substratum interactions during the early stages of melanoma cell spreading on endothelial basement membranes, which is vital for tumor progression. The protein′s structure, characterized by unique chondroitin sulfate glycosaminoglycan chains, allows NG2 to interact with various extracellular matrix components and cell surface receptors, facilitating critical signaling pathways that promote tumor invasion and growth. Notably, phosphorylation at threonine 2256 in rat NG2 (threonine 2252 in human NG2) is essential for redistribution on the surface of astrocytomas, leading to enhanced cell polarization and increased motility, which are key factors in cancer metastasis. Additionally, NG2 serves as a co-receptor for α4β1 integrin, further underscoring its importance in malignant melanoma cell behavior. NG2 expression on blood vessels throughout the rat embryo and on endothelial cells in the central nervous system highlights its significance in vascular development and function. Furthermore, NG2 is recognized as a novel marker for epidermal stem cells, contributing to their organized distribution by promoting clustering, which is essential for maintaining tissue homeostasis and regeneration.
