Neurofibromin Antibody (H-12) is a mouse monoclonal IgG1 kappa light chain antibody that detects Neurofibromin protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-Neurofibromin antibody (H-12) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Neurofibromin, also known as NF1-GAP-related protein, plays a crucial role in regulating cell growth and differentiation by acting as a negative regulator of the RAS signaling pathway, which is vital for normal cellular function and development. Mutations in the NF1 gene can lead to neurofibromatosis type 1, a common autosomal dominant disorder characterized by the development of benign tumors and an increased risk of certain malignancies. Neurofibromin consists of 2,818 amino acids and is expressed consistently across various tissues, including brain, lung, liver, kidney, spleen, muscle, and colon, highlighting its importance in maintaining cellular homeostasis. Structural homology with the catalytic domain of GTPase-activating proteins suggests Neurofibromin may interact with Ras proteins in vivo, further emphasizing its critical role in cellular signaling pathways.

how come the mice anti-NF1 can be used for detection of protein in mice tissue as it is mentioned above in western blot samples for example? thanks

Thank you for your question. This antibody is recommended for detection of Neurofibromin of mouse, rat and human origin. There is representative data in mouse NIH/3T3 whole cell lysate by Western blot. If this does not answer your question, please contact us for further Technical Support.

I would like to use Neurofibromin Antibody (H-12): sc-376886 for double staining with another mouse monoclonal primary antibody. Is this possible?

Rated 5 out of 5 by from Antibody detected neurofibrominNeurofibromin is a very large protein (over 300 kDa) and I tried several antibodies from different vendors in order to obtain a Western image of NF1 in 6 cell types. The Santa Cruz antibody worked very well and detected an appropriately sized protein on PVDF using a TAE 3%-8% gel and Licor IR secondary antibodies. The band is not very prominent but it's there in 5 of 6 cell lines (the 6th was actually expected to NOT have it).

Since this is a mouse monoclonal antibody, we had a lot of trouble with blood vessel staining even after adequate perfusion. The stain definitely worked, however we need to work hard to work around the blood vessels.
