Neomycin solution is a reagent that has been sterile-filtered and made suitable for cell culture. This product is recommended for use in cell culture applications at 5 mL/L. It can be used as a selection agent for prokaryotic cells transformed using the neo selectable marker gene. Also, it is useful for studying ototoxic side effects of antibiotics, platelet-derived growth factor responses in certain fibroblasts, and extraction of Nuclear Phosphatidylinositol 4,5-Bisphosphate-Interacting Proteins. Neomycin′s mechanism of action is to bind to the 30S and 50S subunits, causing miscoding and inhibiting initiation and elongation during protein synthesis. It also blocks voltage-sensitive Ca2+ channels without affecting the Na+/Ca2+ antiporter in neurons. It affects both gram-positive and gram-negative bacteria. Neomycin, derived from Streptomyces fradiae, is a broad spectrum aminoglycoside antibiotic that exhibits antibacterial activity. It is composed of three components, namely isomeric components B and C (active) and neomycin A (minor). By irreversibly binding to the bacterial 30S ribosomal subunit′s 16S rRNA and S12 protein, neomycin disrupts the assembly of the initiation complex between mRNA and the ribosome. This interference inhibits protein synthesis initiation and induces misreading of the mRNA template, leading to translational frameshift and premature termination. Consequently, bacterial cell death occurs.
