NEDD4 Antibody (F-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects NEDD4 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-NEDD4 antibody (F-11) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. NEDD4, a 221 amino acid protein, is primarily located in the Golgi apparatus and is characterized by three transmembrane domains at the carboxy-terminus and two proline-tyrosine (PY) motifs at the amino-terminus. The Golgi localization of NEDD4 plays a significant role in maintaining Golgi structure and function, essential for proper protein sorting and trafficking within cells. NEDD4 interacts with WW domains of several HECT-type E3 ubiquitin ligases, suggesting involvement in ubiquitination processes that regulate protein degradation and cellular signaling pathways. NEDD4 shows high expression in neurons surrounding injury sites, indicating potential neuroprotective functions through ubiquitination modulation, which may contribute to neuronal survival strategies. NEDD4 expression and interaction with E3 ubiquitin ligase Itch increases upon T cell activation, highlighting relevance in immune responses. Mice lacking NEDD4 show impaired Itch activity and Jun B accumulation, leading to severe inflammatory conditions in skin and lungs, resulting in premature mortality.
