Naphthofluorescein inhibits the invasion of Matrigel by the human fibrosarcoma cell line, HT1080. Naphthofluorescein is an inhibitor of Furin. The effects of this furin inhibitor on other furin-mediated processes are still unknown. It has a high affinity for proteins and other molecules, which allows it to bind to them and interact with them. When Naphthofluorescein binds to a molecule, it can alter its structure and function. I am studying on the cleavage of viral envelope glycoproteins by furin and other furin-like proteases. Do this furin inhibitor suppress other furin-like proteases?

Hello. I am conducting research at San Diego State University on how to inhibit MMP-14. Seeing that this substance is activated by Furin, I am interested in Naphthofluorescein. I am curious as to, chemically, how it stops Furin? Thank you for your time.

Thank you for your question. Furin is a proprotein convertase, converting precursor proteins to functional proteins within the Golgi/trans-Golgi secretory pathway. Naphthofluorescein is a cell-permeable inhibitor of furin, it inhibits furin-mediated cleavage of the pro-form of membrane type-1 matrix metalloproteinase (MT1-MMP), resulting in decreased levels of active MT1-MMP.
