N-Hydroxysuccinimide, known as NHS, is a hydroxylated succinimide that is widely used in the activation of carboxylic acids to N-Hydroxysuccinimide-esters to promote coupling with nucleophiles. This approach is most employed in activating the free acid forms of fluorophores for promoting conjugation of these fluorophores to amine residues of proteins. Use of N-Hydroxysuccinimide ester activation has also been described in the synthesis of N-acyl amino acids.
