N-Desmethyl Sildenafil, a metabolite of Sildenafil (sc-338584), derives its significance from its association with Sildenafil, a selective inhibitor of the type 5 cGMP phosphodiesterase (PDE5 inhibitor). PDE5 inhibitors catalyze the hydrolysis of 3′,5′-cyclic guanosine monophosphate (cGMP). Sildenafil, in particular, has demonstrated its potential as a neuro-, synapto-, and angiogenesis enhancer in rat stroke models. Furthermore, it exhibits mild vasodilatory properties. Notably, Sildenafil exerts protective effects against indomethacin-induced small intestinal ulceration through an NO/cGMP-dependent mechanism. Structurally, N-Desmethyl Sildenafil resembles Sildenafil, the active ingredient in the popular erectile dysfunction drug Viagra. Both compounds function as PDE5 inhibitors, impeding the action of phosphodiesterase 5 and consequently elevating cyclic guanosine monophosphate (cGMP) levels. This subsequently augments nitric oxide production. The relaxation of smooth muscle cells in blood vessel walls ensues, leading to improved blood flow. Additionally, N-Desmethyl Sildenafil has demonstrated anti-inflammatory effects and the ability to impede the growth of specific cancer cell types.
