MTHFD1 Antibody (C-3) is a mouse monoclonal IgG1 antibody that detects methylenetetrahydrofolate dehydrogenase 1 (MTHFD1) in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). MTHFD1 is a crucial 935-amino acid, folate-dependent enzyme that facilitates the interconversion of tetrahydrofolate derivatives, which are essential for the synthesis of purines, methionine, and thymidylate, thereby playing a vital role in cellular metabolism and DNA synthesis. MTHFD1 is primarily located in the cytosol and functions as a homodimer, comprising three distinct subunits that contribute to enzymatic activities. The N-terminal domain houses the dehydrogenase and cyclohydrolase activities, while the larger C-terminal domain is responsible for synthetase activity. This structural organization allows MTHFD1 to efficiently catalyze multiple reactions in the folate metabolic pathway, which is critical for cellular proliferation and development. Notably, mutations in the MTHFD1 gene have been linked to an increased risk of neural tube defects in offspring of pregnant women, highlighting MTHFD1′s importance in embryonic development and overall health. MTHFD1′s involvement in folate metabolism has implications in migraine pathophysiology, particularly in cases of migraine with aura, making MTHFD1 a protein of interest in both developmental biology and neurological research.
