MTF-1 Antibody (H-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects MTF-1 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). MTF-1 (H-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The metal-responsive element (MRE)-binding transcription factor, MTF-1, plays a crucial role in cellular response to heavy metal exposure by stimulating metallothionein expression, which is important for metal detoxification and homeostasis. MTF-1 is primarily located in the nucleus, where MTF-1 binds to specific DNA sequences to regulate gene expression. Nuclear localization allows MTF-1 to effectively respond to changes in metal ion concentrations, thereby influencing cellular stress responses and maintaining metal ion balance. MTF-1′s structure includes six zinc fingers in its DNA-binding domain, which are essential for interaction with target genes. Phosphorylation of MTF-1 upon metal exposure enhances transcriptional activity, further underscoring significance in cellular adaptation to environmental stressors. Beyond metallothionein activation, MTF-1 participates in a post-transcriptional regulatory complex that modulates ribosomal protein S25 mRNA export, highlighting MTF-1′s multifaceted role in cellular regulation.

Rated 4 out of 5 by from Good antibodyI used the free trial sample for the WB (around 30 ug total protein for each lane; blocking in 5% BSA TBST; overnight 4'C, primary 1:100 dilution in 5%BSA TBST; secondary: 1:1000, room temperature for 1 hour). MDA-MB-231 cells were used for my experiment, in general, this antibody can recognize MTF-1 (around 130kDa) as shown by nucleus translocation in the presence of ZnCl2 or siRNA knockdown. (although the background is a little high, and some non-specific bands are observed, maybe different blocking reagent or changing the dilution of primary or secondary antibody will help to resolve this issue)
