ML-180 is an antagonist for liver receptor homolog-1 (LRH1), also known as Nuclear Receptor Subfamily 5 Group A Member 2 (NR5A2). LRH1 is a transcription factor that plays a role in various physiological processes, including liver development, bile acid metabolism, steroidogenesis, and regulation of lipid and glucose homeostasis. ML-180 is inactive at the related steroidogenic factor-1 transcriptional activator. Through LRH1, ML-180 alters the expression of haptoglobin and serum amyloid proteins A1 and A4, induces the death of estrogen-receptor negative MDA-MB-231 breast cancer cells, and inhibits the steroidogenic acute regulatory promoter.
