Meclofenamate sodium is a dual inhibitor of the 5-LO (5-lipoxygenase) and Cox (cyclooxygenase) pathways. Meclofenamate sodium is demonstrated to inhibit both Cox-1 and Cox-2, showing micromolar inhibition of human recombinant hCox-1 (IC50 of 1.5 muM) and hCox-2 (IC50 of 9.7 muM). An irreversible time-dependent inhibition of hCOX-2 is described, involving a rapid equilibrium binding of enzyme and inhibitor followed by formation of a tightly bound inhibited complex.
