Ly-6G Antibody (RB6-8C5) is a rat monoclonal IgG2b antibody that detects the Ly-6G protein of mouse origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). Ly-6G (RB6-8C5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The Ly-6G protein, also known as Lymphocyte antigen 6G, is a member of the Ly-6 family of glycosyl-phosphatidylinositol (GPI)-anchored proteins, characterized by a distinctive cysteine-rich domain that contains O-linked carbohydrates. Ly-6G protein is predominantly expressed on neutrophils, where Ly-6G localizes to the cell membrane, playing a crucial role in the immune response. Notably, Ly-6G is part of the Gr-1 antigen complex, which is expressed on monocytes, neutrophils, and a subset of macrophages, as well as plasmacytoid dendritic cells and T cells. Ly-6G expression on these immune cells is significant as Ly-6G is involved in the regulation of immune responses, including the suppression of antitumor activity and T cell responses, making Ly-6G a valuable marker for studying neutrophil biology and their role in various pathological conditions.

Will you please provide me with the protocol used to run the western blot for the Ly-6G Antibody (RB6-8C5): sc-53515 that was performed under non-reducing conditions. It would be most helpful to know the composition of the Transfer Buffer. Thank you.

Thank you for your question. We followed our recommended protocol for Western blotting. Except, for the mouse PBL lysate we added our non-reducing electrophoresis buffer (sc-45085), and loaded on the regular SDS-PAGE gel without boiling. For our transfer buffer, we use sc-363874 Towbin powder, and add methanol at 10%. The bottle of Towbin powder makes 20L of 1x.

How can Ly-6G (RB6-8C5): sc-53515 mouse monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This antibody is available conjugated to phycoerythrin (sc-53515 PE) or fluoroscein (sc-53515 FITC).
