LHX3 Antibody (2C10) is a mouse monoclonal IgG2a kappa light chain antibody that detects LHX3 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). LHX3 (2C10) antibody is available as the non-conjugated anti-LHX3 monoclonal isotype antibody. LHX3 protein plays a crucial role in the development of the central nervous system, particularly in the specification of motor neurons and the formation of the pituitary gland. LHX3 is essential for regulating cell migration and axon navigation during neuronal development. LHX3 exists in two isoforms, LHX3a and LHX3b, which differ at the amino terminus but share conserved LIM domains and a homeodomain that is vital for function. The homeodomain contains three nuclear localization signals, allowing LHX3 to effectively target the nucleus, where LHX3 exerts transcriptional regulatory functions. Both isoforms are predominantly expressed in the adult pituitary gland, spinal cord, and lungs, highlighting their importance in these tissues. The short LHX3b isoform has been shown to inhibit DNA binding and transcriptional activity, indicating a complex regulatory mechanism. Human LHX3 is located at the subtelomeric region of chromosome 9 at band 9q34.3, a site associated with chromosomal translocations and insertions, suggesting involvement in various developmental disorders of the central nervous system.

Can sc-293411: LHX3 (2C10) monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

    Asked by: TinTin
Thank you for your question. Yes, sc-293411: LHX3 (2C10) is recommended for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
