LEKTI Antibody (E-9) is a mouse monoclonal IgG2a kappa light chain antibody that detects LEKTI protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). LEKTI (E-9) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The lympho-epithelial Kazal-type inhibitor (LEKTI) plays a crucial role as a serine protease inhibitor, providing essential protection to mucous epithelia against microbial invasion and inflammation, which is vital for maintaining the integrity of epithelial barriers. LEKTI is predominantly located in the granular and uppermost spinous layers of the epidermis, as well as in the differentiated layers of stratified epithelia, highlighting LEKTI′s importance in epithelial differentiation and function. Notably, mutations in the SPINK5 gene, which encodes LEKTI, are linked to Netherton syndrome, a severe autosomal recessive disorder characterized by atopic dermatitis, hay fever, and other related conditions, underscoring LEKTI′s significance in skin health and disease.

1-Would be thankful to let me know which coating solution would be the best to use for sitting the E-9 antibody on the plate as the capture AB in the sandwich Elisa. 2- What is the recommended capture and detection concentration in Elisa for this AB?

What is the recommend dilution of the Ab for WB? Is this Ab was used for WB application in published works?

Thank you for your question. For this antibody, the recommended starting dilution is 1:100, with a dilution range between 1:100-1:1000. as of yet, there are no publications citing use of this antibody in western blots. Anti-LEKTI Antibody (E-9) is recommended for detection of LEKTI precursor and mature form and HF7665 active peptide of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA

Does the unmasking have to be performed with citrate or EDTA for IHC on paraffin-embedded tissues ?

Thank you for your question. You can find our recommended protocol by following this link: https://www.scbt.com/de/resources/protocols/immunoperoxidase-staining. We recommend Heat treatment: Place slides in a container and cover with 10 mM sodium citrate buffer, pH 6.0 (sc-294091); or with 50 mM glycine-HCl buffer (sc-29096), pH 3.5, with 0.01% (w/v) EDTA (sc-29092). Heat at 95° C for 5 minutes. Top off with fresh buffer and heat at 95° C for 5 minutes (optimal incubation time may vary for each tissue type). Allow slides to cool in the buffer for approximately 20 minutes. Wash in deionized H2O three times for 2 minutes each. Aspirate excess liquid from slides.

I would like to know if this antibody has been tested for IHC in FFPE sections of mouse epidermis.

Thank you for your question. This antibody has been tested for IHC in FFPE tissue sections, or for IHC(P) in human tonsil. We have not tested this in mouse epidermis.

What is the recommended fixation method for immunofluorescence staining with LEKTI (E-9): sc-137109 monoclonal antibody?
