LARS2, also known as leucyl-tRNA synthetase 2, mitochondrial or LEURS, is a crucial 903 amino acid protein belonging to the class-I aminoacyl-tRNA synthetase family, primarily localized in the mitochondrial matrix. LARS2 plays a vital role in protein synthesis by catalyzing the aminoacylation of leucine to tRNA(Leu) through a two-step reaction, which begins with the activation of leucine by ATP to form an adenylate intermediate. This intermediate then transfers the activated leucine to the 3′-end of the target tRNA, a process essential for accurate translation and protein formation. The specific sequence and orientation of the variable loop in tRNA(Leu) are critical for interaction with LARS2, ensuring the fidelity of protein synthesis. LARS2 has been implicated in various neurological and metabolic disorders, including bipolar disorder and schizophrenia, where upregulation may lead to the production of a mutated tRNA(Leu) that attempts to undergo aminoacylation.
