kynureninase Antibody (E-5) is a mouse monoclonal IgG1 kappa light chain antibody that detects kynureninase protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). kynureninase (E-5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Kynureninase, also known as L-kynurenine hydrolase, is a 465 amino acid enzyme predominantly located in the cytoplasm, where kynureninase plays a crucial role in tryptophan metabolism. This enzyme is essential for L-kynurenine degradation and NAD(+) cofactor biosynthesis, catalyzing L-kynurenine cleavage into anthranilic acid and L-3-hydroxykynurenine into 3-hydroxyanthranilic acid, with a notable preference for the L-3-hydroxy form. Kynureninase forms a homodimer structure that enhances stability and functionality, allowing for efficient substrate processing. Additionally, kynureninase utilizes pyridoxal phosphate as a cofactor and is inhibited by o-methoxybenzoylalanine (OMBA). Kynureninase is widely expressed in various tissues, with highest levels found in lung, placenta, and liver. A kynureninase deficiency can lead to hyperkynureninuria, a metabolic disorder characterized by impaired conversion of tryptophan to nicotinic acid (vitamin B6), while increased kynureninase activity has been associated with systemic and cerebral inflammatory conditions, highlighting kynureninase′s potential role in disease states.
